Sterile Preparation2

II) Ophthalmic preparation:
These are sterile preparation which is applied to the eye in the form of drops, ointment, suspension or ocuserts.
Sterilization the ophthalmic preparation by:

1.  Dissolved active constituent in suitable solvent which contain antimicrobial agent → filter→ trans to final container & firm close → sterilized in autoclave (121  for 20min.)
2.  The same later step with sterilized & firm close without use autoclave.
3.  The same later step without use autoclave but storage at 98  → 100  for 30min.

Why Sterilization the ophthalmic preparation?
Because pseudomonas aeruginosa → corneal ulceration → blindness.

The preservative used in ophthalmic preparation:
1.  Benzalkonium: has broad spectrum but not effecting against pseudomonas aeruginosa so used with polymycinB.
2.  Phenyl mercuric nitrate: it ppt on the eye lens if it is used for long time so don't used with glaucoma.

Packaging of ophthalmic solution & suspension: in an opaque flexible plastic container made of nylon fitted with a fixed or building dropper.

Packaging of ophthalmic ointment: into a collapsible tin or plastic tube which closed with screw caps which made of aluminum or plastics.

Ocuserts: a newly device for the insertion of the drug to the lower eye lid which provide a constant and continues drug release for long period of time.
